Abstract
The OrganoScreen platform aims to provide preclinical tumoroid models based on 3D culture of tumor cells harvested from patients. We are working to establish an institutional biobank that will enable us to 1) explore the mechanisms of resistance to pharmacological and immunotherapeutic treatments used to fight cancer and 2) discover new therapeutic targets to offer patients more personalized medicine.
To achieve this, we draw on two areas of expertise: 1) the generation of multi-organ patient-derived organoids (PDOs), potentially incorporating stromal and immune cells into microfluidic models, and 2) drug screening or genetic screening using CRISPR/Cas9 technology.
Services offered by the platform: development of new models, biobanking, drug screening, CRISPR screening, and R&D.

IHU PRISM: Oasis, Unlock, Meditwin (multi-organ).
Panoramic Cohort: Pancreatic ductal adenocarcinomas (PDAC).
RHU Organomic: PDAC and colorectal cancer (CRC).
SIRIC: Sarcomas.

Services Offered
The platform supports its employees throughout the experimental process. We define the scope of projects during a kickoff meeting and then prepare a preliminary quote for approval. Here is the list of services offered:
1) Development of multi-organ 3D models from tumors: testing of media and matrices for the generation of relevant preclinical models.
2) Biobanking: storage of cell lines following molecular characterization (RNA-seq, WES) and imaging (EVOS5000, confocal, and Tomocube).
3) Drug screening: IC50 (kill curve), automated MTG viability assay (TECAN, Biotek Synergy LX)
4) CRISPR screening: on 3D models or cell lines. Design of a CRISPR library for bulk KO applications. Analysis of results via NGS.
5) R&D: microfluidics (CherryBiotech), co-culture, miniaturization (Okomera), and automation.
